This study analyzes the effects of the educational game Nucleus Quest on the teaching of cytology in high school, considering both students’ conceptual performance and their perceptions of the learning experience. The methodological design was quasi-experimental, involving the administration of pre- and post-tests to 394 students from a public school in the northern region of Brazil. The game, designed as a multimedia board, was structured in gradual challenges that required cooperation, decision-making, and conceptual mastery. Statistical analysis showed significant differences between pre- and post-test results, indicating learning improvement. In addition, the perception questionnaires revealed high acceptance of the proposal, especially regarding content relevance, clarity of the rules, and contribution to conceptual understanding, although aspects related to social interaction received less favorable evaluations. The findings suggest that the use of games in the classroom, when intentionally integrated into the curriculum, is an effective strategy to promote engagement and learning gains in biology, in alignment with the competencies established by the BNCC. It is concluded that the educational game Nucleus Quest represents an innovative pedagogical resource with the potential to strengthen the articulation between theory and practice in science education.
